If the sig radius of your target is large enough and you're shooting in to the fall-off range of your weapon, will the sigradius negate the -ve to hit modifier that is applied because of falloff? I.e. will you hit at 100% even though you're in falloff range?

Nope. The sig radius of your target can have a large effect on your hit chance at short ranges. In falloff range the effect is slighty lower.
Take this example:
A Taranis fitted with 125mm Railguns II and Antimatter S ammo orbiting your target with a transversal speed of 500 m/s:
- Tracking: 0,14609 - Optimal range: 6,75 km - Falloff: 6,25 km - Sig resolution gun: 40
See the graph for 4 different targets: - Frigate: sig radius 40 - Cruiser: sig radius 135 - Battlecruiser: sig radius 240 - Battleship: sig radius 370
Hit Chance Graph
Notice how a bigger sig radius of your target helps you improve your hit chance at short ranges, where the tracking of your guns against a frigate was an issue. At ranges above optimal the difference is minimal.

Ermm... Excel.
Let me rephrase for clarity:
At short ranges, where your guns could have tracking issues, shooting a bigger target (higher sig radius) will improve your hit chance considerably.
Above optimal range, having a bigger target won't negate the falloff penalty, but it can offer a slightly better hit chance.

Try the Eve Dev Wiki tracking formula page
You can also generate a DPS chart using EFT's DPS graphs. File -> New DPS Graph.
